mtDNA RFLP to catch a cat burglar

This practical was designed for 2nd year undergraduate students as part of a Genetics module. They were given lectures on the existence, structure and inheritance patterns of mitochondrial DNA (mtDNA). They were also shown a paper by Giles et al. (1980), that demonstrated the maternal inheritance of human mtDNA using RFLP analysis. Students would also need to be familiar with the use of restriction digests, gel electrophoresis and PCR.

It runs over one computer-based planning session followed by two lab-based sessions.
